We are delighted to announce that the 2011 training workshop on nonhuman primate MHC genotyping by Roche/454 pyrosequencing will be held in Madison, WI the week of November 7-12. This workshop is a component of the NIH NIAID contract "Nonhuman Primate Major Histocompatibility Complex Gene Discovery and Typing Technology Development. (HHSN272201100013C).
The workshop provides training in all aspects of pyrosequencing-based MHC class I genotyping, including:
• isolation of RNA from peripheral blood mononuclear cells
• preparation of first-strand cDNA
• generation of primary cDNA PCR amplicons
• automated and manual cleanup of primary cDNA PCR amplicons
• quantification of PCR amplicons
• preparation of multiplexed PCR amplicon pools for pyrosequencing
• emulsion PCR preparation
• breaking emulsion PCRs and enriching library beads
• loading and running the Roche/454 GS Junior pyrosequencer
• analysis of GS Junior pyrosequencing data with Geneious Pro
